Historical Developments

1910Stéphane Leduc published the first identifiable use of the term 'synthetic biology' 116 years ago

Leduc used the phrase in Théorie physico-chimique de la vie et générations spontanées, and again in La Biologie Synthétique in 1912. This gave the field its eventual name decades before the molecular tools to realise it existed.

1944Oswald Avery showed that DNA is the material of genes and chromosomes 82 years ago

The Canadian-American scientist's demonstration established DNA as the hereditary molecule. It became the bedrock on which all subsequent genetic research, and later synthetic biology, was built.

1953Francis Crick and James Watson published the structure of DNA in Nature 73 years ago

The double-helix structure revealed how genetic information is encoded and copied. It made rational manipulation of genetic sequence conceivable.

1961François Jacob and Jacques Monod described the operon model of gene regulation 65 years ago

Their study of the lac operon in E. coli established that bacteria switch genes on and off in response to their environment through molecular circuitry, giving biology its first account of genetic regulatory logic. They explicitly envisioned assembling new systems from molecular components, anticipating the engineering logic later synthetic circuits such as the repressilator were built to exploit.

1973Cohen, Boyer et al. published the first molecular cloning and amplification of DNA in a plasmid 53 years ago

The P.N.A.S. paper demonstrated that DNA could be recombined and propagated in a host organism. It is widely regarded as the dawn of synthetic biology.

1976Har Gobind Khorana's laboratory chemically synthesised the first artificial gene shown to function inside a living cell 50 years ago

The Indian-born biochemist's team built a yeast transfer RNA gene from individual chemical building blocks and showed it worked once inserted into bacteria, establishing that genes could be constructed rather than only extracted from existing organisms.

1978Arber, Nathans and Smith won the Nobel Prize for the discovery of restriction enzymes 48 years ago

Restriction nucleases permitted easy construction of recombinant DNA molecules and analysis of individual genes. Szybalski's editorial in Gene marked the arrival of a 'new era of synthetic biology' in which new gene arrangements could be constructed and evaluated.

1988Mullis et al. published PCR using a thermostable DNA polymerase 38 years ago

The Science paper removed the need to add fresh polymerase after each cycle of the polymerase chain reaction. This greatly simplified DNA mutagenesis and assembly, making routine construction of designed sequences practical.

1991James Bailey defined metabolic engineering as a distinct discipline 35 years ago

His paper set out the deliberate redesign of an organism's metabolic pathways using recombinant DNA methods as a systematic engineering discipline rather than ad hoc genetic modification, a framing that underpins the industrial strain design synthetic biology later built on.

2000Michael Elowitz and Stanislas Leibler, and separately Timothy Gardner, Charles Cantor and James Collins built the first synthetic genetic circuits 26 years ago

The repressilator and the genetic toggle switch wired existing genes into bacteria to oscillate or flip between states like simple electronic circuits, demonstrating that genetic regulation could be engineered predictably and marking most historians' starting point for synthetic biology as a field.

2000Researchers synthesised the 9.6 kbp Hepatitis C virus genome from chemical oligonucleotides 26 years ago

2003David Baker's laboratory designed Top7, a protein with a structure not found in nature 23 years ago

Using computational modelling rather than modifying an existing natural protein, the team designed an entirely new protein fold from scratch and had it made and shown to fold as predicted, a foundational result for the later field of de novo protein design.

2003Researchers assembled the bacteriophage Phi X 174 genome in about two weeks 23 years ago

The 5386 bp genome was built far faster than the poliovirus effort. The speed increase signalled that genome synthesis was becoming a routine engineering operation.

2010Craig Venter's team created the first cell controlled entirely by a chemically synthesised genome 16 years ago

Researchers synthesised the genome of Mycoplasma mycoides, JCVI-syn1.0, from digital DNA sequence data, assembling chemically synthesised fragments by yeast recombination and transplanting the result into a genomically emptied cell of a related species, which then began replicating under the synthetic genome's control. Widely described as creating the first synthetic life, it was the field's landmark demonstration that an entire functioning genome could be built to design.

US President Barack Obama asked a bioethics commission to review the implications within days of the announcement
December 2010US Presidential Commission for the Study of Bioethical Issues issued the report New Directions: The Ethics of Synthetic Biology 16 years ago
